Proteins that work at the ends of DNA could provide cancer insight

ScienceDaily (Nov. 29, 2012) ? New insights into a protein complex that regulates the very tips of chromosomes could improve methods of screening anti-cancer drugs.

Led by bioengineering professor Sua Myong, the research group's findings are published in the journal Structure.

Myong's group focused on understanding the proteins that protect and regulate telomeres, segments of repeating DNA units that cap the ends of chromosomes. Telomeres protect the important gene-coding sections of DNA from loss or damage, the genetic equivalent of aglets -- the covering at the tips of shoelaces that keep the ends of the laces from unraveling or fraying.

Telomeres play an important role in cell aging and death, since each time a cell divides, a little bit is lost from the end of the telomere. Thus, cell biologists postulate that telomere length can determine the lifespan of a cell. Cancer cells, however, have a way to get around this limitation: An enzyme called telomerase that adds length to telomeres is highly active in cancer cells. This allows cancer cells to divide in perpetuity, running amok through tissues and systems.

"Cancer researchers want to get a hold of this problem, control this indefinite lengthening of the telomeres," said Myong, who also is affiliated with the Institute for Genomic Biology at the U. of I. "A lot of the anti-cancer drugs are targeted directly to these telomeres so that they can inhibit telomerase activity. The proteins we study regulate the activity of telomerase."

Using a technique developed at Illinois that allows researchers to watch single molecules interact in real time, Myong's group determined how two proteins called POT-1 and TTP-1 bind to the telomere. POT-1 protects the fragile telomere ends from being attacked by other regulatory proteins that might mistake the end for a broken or damaged area of DNA. When POT-1 and TTP-1 work together in a complex, they promote telomerase activity, an interesting target for cancer researchers.

The group found that on its own, POT-1 binds to the folded-up telomere in distinct steps at particular points in the telomere's DNA sequence, unfolding the telomere in a stepwise manner. However, the POT-1/TTP-1 complex surprised the researchers by binding, then freely sliding back and forth along the telomere end.

"Instead of stepwise binding, what we saw was a mobile protein complex, a dynamic sliding motion," Myong said. "Somehow it was as if the static binding activity of POT-1 is completely lost -- the protein complex just slid back and forth. We were able to reproduce the data and confirm it with many different tail lengths of the telomeric DNA and we know now that the contact between POT-1 and the telomere is somehow altered when the partner protein comes and binds."

Next, the researchers will add telomerase and see how the sliding activity of the POT-1/TTP-1 complex affects telomerase activity. Myong postulates that the sliding may promote telomerase activity -- and thus telomere lengthening -- by making the end of the telomere accessible for the telomerase enzyme to bind.

"We are excited about the possibility that this kind of mobility can increase the telomerase extension activity," Myong said. "It's somehow engaging the enzyme so that it can stay bound to the DNA longer. So it must involve a direct interaction."

Ultimately, understanding the POT-1/TTP-1 complex gives drug developers a new target for anti-cancer drugs, and the assay Myong's group used to monitor the complex could offer a venue for evaluating telomere-targeting drugs.

"We want to extend our a basic science knowledge in telomere biology into causes of cancer and we hope that our assay can be useful for telomere-targeted drug screening," Myong said.

Precisely engineering 3-D brain tissues

ScienceDaily (Nov. 29, 2012) ? Borrowing from microfabrication techniques used in the semiconductor industry, MIT and Harvard Medical School (HMS) engineers have developed a simple and inexpensive way to create three-dimensional brain tissues in a lab dish.

The new technique yields tissue constructs that closely mimic the cellular composition of those in the living brain, allowing scientists to study how neurons form connections and to predict how cells from individual patients might respond to different drugs. The work also paves the way for developing bioengineered implants to replace damaged tissue for organ systems, according to the researchers.

"We think that by bringing this kind of control and manipulation into neurobiology, we can investigate many different directions," says Utkan Demirci, an assistant professor in the Harvard-MIT Division of Health Sciences and Technology (HST).

Demirci and Ed Boyden, associate professor of biological engineering and brain and cognitive sciences at MIT's Media Lab and McGovern Institute, are senior authors of a paper describing the new technique, which appears in the Nov. 27 online edition of the journal Advanced Materials. The paper's lead author is Umut Gurkan, a postdoc at HST, Harvard Medical School and Brigham and Women's Hospital.

'Unique challenges'

Although researchers have had some success growing artificial tissues such as liver or kidney, "the brain presents some unique challenges," Boyden says. "One of the challenges is the incredible spatial heterogeneity. There are so many kinds of cells, and they have such intricate wiring."

Brain tissue includes many types of neurons, including inhibitory and excitatory neurons, as well as supportive cells such as glial cells. All of these cells occur at specific ratios and in specific locations.

To mimic this architectural complexity in their engineered tissues, the researchers embedded a mixture of brain cells taken from the primary cortex of rats into sheets of hydrogel. They also included components of the extracellular matrix, which provides structural support and helps regulate cell behavior.

Those sheets were then stacked in layers, which can be sealed together using light to crosslink hydrogels. By covering layers of gels with plastic photomasks of varying shapes, the researchers could control how much of the gel was exposed to light, thus controlling the 3-D shape of the multilayer tissue construct.

This type of photolithography is also used to build integrated circuits onto semiconductors -- a process that requires a photomask aligner machine, which costs tens of thousands of dollars. However, the team developed a much less expensive way to assemble tissues using masks made from sheets of plastic, similar to overhead transparencies, held in place with alignment pins.

The tissue cubes can be made with a precision of 10 microns, comparable to the size of a single cell body. At the other end of the spectrum, the researchers are aiming to create a cubic millimeter of brain tissue with 100,000 cells and 900 million connections.

Answering fundamental questions

Because the tissues include a diverse repertoire of brain cells, occurring in the same ratios as they do in natural brain tissue, they could be used to study how neurons form the connections that allow them to communicate with each other.

"In the short term, there's a lot of fundamental questions you can answer about how cells interact with each other and respond to environmental cues," Boyden says.

As a first step, the researchers used these tissue constructs to study how a neuron's environment might constrain its growth. To do this, they placed single neurons in gel cubes of different sizes, then measured the cells' neurites, long extensions that neurons use to communicate with other cells. It turns out that under these conditions, neurons get "claustrophobic," Demirci says. "In small gels, they don't necessarily send out as long neurites as they would in a five-times-larger gel."

In the long term, the researchers hope to gain a better understanding of how to design tissue implants that could be used to replace damaged tissue in patients. Much research has been done in this area, but it has been difficult to figure out whether the new tissues are correctly wiring up with existing tissue and exchanging the right kinds of information.

Another long-term goal is using the tissues for personalized medicine. One day, doctors may be able to take cells from a patient with a neurological disorder and transform them into induced pluripotent stem cells, then induce these constructs to grow into neurons in a lab dish. By exposing these tissues to many possible drugs, "you might be able to figure out if a drug would benefit that person without having to spend years giving them lots of different drugs," Boyden says.

Weak start to November hits some big U.S. retailers

(Reuters) - Leading U.S. retailers said weak sales in early November, when people in the Northeast were cleaning up after superstorm Sandy, dragged down results for the month, despite brisk activity over the long Thanksgiving weekend.

Target Corp and Macy's Inc, two of the biggest companies to report monthly sales on Thursday, missed analysts' expectations. Both said the weakness came at the beginning of November, not during the Thanksgiving weekend rush.

Nordstrom Inc also blamed its disappointing November sales on a lackluster first half of the month, both because of Sandy and a poor showing at its sale for women and kids.

Overall, retailers on average reported a 1.6 percent increase in sales at stores open at least a year, about half the 3.3 percent rise that analysts had forecast, according to Thomson Reuters I/B/E/S. The results include data from 16 retailers, but not drugstores. Zumiez Inc reports later on Thursday.

Same-store sales at the chains that report them are closely watched for indications of consumer spending during the beginning of the holiday season. Analysts had not expected the November results to match the 3.5 percent gain of a year earlier.

Discounts reign during the holiday season, with stores hopeful that customers who come in for deals such as Thanksgiving weekend "doorbusters" also pick up items that are not heavily marked down.

Madison Riley, managing director at retail consulting firm Kurt Salmon, said there had been "a lot of very heavy promotions" during Thanksgiving weekend.

"I think a lot of that consumer activity was focused on the doorbusters, and I don't know how much it broadened into the full store assortment," Riley said. "It may be a bit of a squeeze from a margin standpoint."

Fifty-two percent of Black Friday shoppers who participated in a Reuters/Ipsos poll said they had stayed on budget, and 34 percent said they had spent less than planned. Only 14 percent said they had gone over budget.

Despite the largest-volume Thanksgiving weekend in Macy's Inc's history, the department store operator was not able to overcome the weak start of November, Chief Executive Officer Terry Lundgren said in a statement.

While Macy's posted an unexpected same-store sales decline, the company is on track for a "very strong" performance in the current, holiday quarter, Lundgren said.

Target's 1 percent decline in same-store sales stemmed from weakness in the first two weeks of November, CEO Gregg Steinhafel said in a statement. Sales picked up later in the month, and profitability was as planned, Steinhafel added.

Limited Brands Inc said its same-store sales had risen 5 percent, including a hit of one to two percentage points from Sandy. The results topped analysts' expectations for a 3.1 percent increase.

Gap Inc's comparable sales rose 3 percent, while Wall Street was expecting a rise of 3.9 percent. However, all of the company's brands did better than last year, led by a turnaround at Old Navy, which lured shoppers on Black Friday with items such as $15 jeans and $5 fleece jackets.

HOLIDAY OUTLOOKS

Fitch Ratings said on Wednesday that it expected 2012 holiday retail sales to grow between 3 percent and 4 percent, down from a 5.6 percent increase in 2011, even though the season got off to a strong start.

The National Retail Federation still expects sales in November and December to rise 4.1 percent this year, below last year's 5.6 percent increase. Total spending was up 12.8 percent at $59.1 billion over the long Thanksgiving weekend, according to an NRF survey.

Cyber Monday deals are not a factor in retailers' November reports, as most chains are reporting their sales through Saturday, November 24. Cyber Monday, the Monday after Thanksgiving that this year fell on November 26, was reportedly the biggest online shopping day ever.

Some retailers that analysts have said did well over the weekend, such as Wal-Mart Stores Inc and online leader Amazon.com Inc, do not report monthly sales.

Same-store sales at Costco Wholesale Corp, the biggest chain that issues monthly reports, rose a better-than-expected 6 percent, the warehouse club operator said on Wednesday.

Fujistu spotted this gap, and created Wandant -- a sensor-laden neck tag for dogs complete with cloud-based service for tracking your pooch's wellbeing. The hardware will log your pet's footsteps, amount of shivering plus temperature change, and upload it automatically via your smartphone or PC. The software will present you these data over time, with the option of manually inputting further details such as food amount, a diary with photos and -- for those with stronger constitutions -- stool condition. Like human-focused equivalents, there will also be provisions for sharing your dog's efforts, but thankfully only with vets and research institutions. There's no details on hardware pricing just yet, but the online component is free for the first year (JPY 420 / about $5 per month there after). Money no object for your mutt? Sadly only available in Japan for the time being, so you'll need to include a return ticket in that splurge too.

Smartphone Study is Good News for Apple, Bad News for Canon

Apple (NASDAQ: AAPL), Samsung, Nokia (NYSE: NOK), Research In Motion (NASDAQ: RIMM) and other smartphone manufacturers can rejoice in the news that consumers are repeatedly turning to their cellular devices to perform a wide array of tasks.

According to PCMag, 82 percent of smartphone users turn on the device just to take a picture, while 80 percent turn it on to send or receive a text message. More than half (56 percent) turn it on to surf the Web; 50 percent do so to send or receive an e-mail.

These findings are not as surprising as they are damaging to companies that produce specific-use devices. Canon (NYSE: CAJ), for example, does not make cellular devices. But it produces a number of digital cameras, including several pocket-sized devices. If Consumers are reaching for their smartphones instead of an actual camera to take photos, Canon's future could be in jeopardy.

Year-to-date Canon shares are down more than 22 percent. The company experienced a few spikes during the summer months, gaining more than 12 percent. Those gains were all but diminished as investors abandoned the stock at the end of the season. They came back in September, raising Canon shares by 14 percent. Those gains were short-lived, however. In the weeks that followed, the company lost more than 17 percent of its value.

Up until this week, Canon had been experiencing another spike, rallying 15 percent since November 14. Those gains have not been able to offset the company's overall losses, however.

At the same time, Apple's shares have been indirectly enhanced by the camera featured in the iPhone. While no single stock move can be attributed to its photographic capabilities, it is no secret that millions of consumers purchased the iPhone 5 for its camera. Despite having endured a three-month decline of more than 13 percent, Apple shares are still up more than 42 percent year-to-date.

(AP) ? The garment factory in Bangladesh where 112 people were killed in a fire had been making clothes for Wal-Mart, Disney, Sears and other major global retailers ? some of whom say they thought they had stopped doing business with the place.

The apparent confusion underscored what some industry experts say is a major obstacle to improving factory safety in developing nations: Many retailers in the U.S. and Europe rely on such a long and complex chain of manufacturers, vendors and middlemen to keep their shelves stocked that it is difficult to keep track of where certain products are made.

Amid the blackened tables and melted sewing machines at Tazreen Fashions Ltd., an Associated Press reporter discovered clothes and account books Wednesday that indicated the factory was used by a host of U.S. and European retailers.

Among the items discovered: children's shorts with Wal-Mart's Faded Glory label, hooded sweaters marked "Disney Pixar," shorts with hip-hop star Sean Combs' ENYCE tag, and sweaters from the French company Teddy Smith and the Scottish company Edinburgh Woollen Mill. Sears was among the companies listed in the account books.

The tragedy is putting a spotlight on dangerous workplace conditions around the world, with no clear answers to how consumers should react or who is ultimately responsible.

Wal-Mart said that it received a safety audit that showed the factory was "high-risk" and had decided well before the blaze to stop doing business with Tazreen. But it said a supplier had continued to use Tazreen without authorization.

When pressed for an explanation of how a supplier could use a factory without the retailer's approval and whether it happened often, Kevin Gardner, a Wal-Mart spokesman, did not directly address the issue in emails to The Associated Press.

Sears said it learned after the blaze that its merchandise was being produced there without its approval through a vendor, which has since been fired. Walt Disney Co., which licenses its characters to clothing makers, said its records indicate that none of its licensees have been permitted to make Disney-brand products at the factory for at least a year.

Retailers like Wal-Mart have contract clauses that require suppliers to disclose all factories and subcontractors producing merchandise for sale. But it's hard to crack down on unauthorized subcontracting, said Josh Green, chief executive of New York-based Panjiva, which tracks shipments for factories outside the U.S.

"The reality is you have to have round-the-clock monitoring of every aspect of the supplier's operations," he said. "It's just not feasible."

Green noted that subcontracting is pervasive as suppliers look for ways to cut costs.

"You have relentless pressure that consumers put on retailers and that retailers put on their suppliers to deliver lower and lower prices," he said. "And that pressure is a key reason why you see factories cutting corners."

Bangladesh's fast-growing garment industry ? second only to China's in exports ? has long provided jobs and revenue for the desperately poor country, while turning out the low-priced products shoppers in the U.S. and other countries have come to enjoy. But the industry has a ghastly safety record; more than 300 workers have died in garment factory fires in Bangladesh since 2006.

Labor activists have seized on the blaze ? the deadliest in Bangladesh's nearly 35-year history of exporting clothing ? to argue that retailers must insist on more stringent fire standards.

Charles Kernaghan, director of the Institute for Global Labour and Human Rights, said nothing will change unless clothing companies protect workers as vigorously as they do their brands.

"The labels are legally protected," he said. "But there are no similar laws to protect rights of the worker."

Green said some companies are more conscientious than others in selecting factories. Some pick a manufacturer and do little or no investigation, he said, while others analyze factories' past infractions and pay monthly visits.

"There's a fundamental difference between companies who do the check-the-box approach and companies who are serious about due diligence," he said.

On Wednesday, police arrested three factory officials suspected of locking in the workers who died in Saturday's blaze on the outskirts of Dhaka. Police Chief Habibur Rahman said the three will be questioned. He said the factory owner was not among those arrested.

About 1,400 people worked at the factory, about 70 percent of them women. Survivors said exit doors were locked, and a fire official said the death toll would have been much lower if the eight-story building had had an emergency exit.

The fire broke out on the ground floor, where a factory worker named Nasima said stacks of yarn and clothes blocked part of the stairway. Nasima, who uses only one name, and other workers said that when they tried to flee, managers told them to go back to their work stations.

"Everyone was screaming for help," Nasima said. "Total chaos, panic and screaming. Everyone was trying to escape and come out. I was pulling the shirt of a man. I fainted and when I woke up I found myself lying on the road outside the factory. I don't know how I survived."

Workers expressed support for the factory owner, Delwar Hossain. Mohammad Rajib said he is "a gentle man" who heeded workers when they protested for more pay and against rough treatment by some managers.

"He took action and fired some of them," he said. "He did not sack any worker. He told us: 'You are my people. If you survive, I will survive.'"

"They were just kind of chilling," freelance photographer Will Byington said. "They were hanging out and not even doing much. They were kind of just checking out the scene on a Friday night in Wrigleyville. It was like they were on a date, taking a stroll."

Byington said he was shooting a concert at a bar across the street from the stadium in the Wrigleyville neighborhood, notoriously crowded and often rowdy on weekend nights, when he saw the two coyotes hanging out by the statute of former Chicago Cubs player Ernie Banks, near the ticketing area.

"It was kind of like they were looking for tickets," the 34-year-old Chicago resident laughed. "They went by the ticket window and unfortunately found it was closed, so they were ready to move onto the bar."

Wildlife ecologist Stanley Gehrt, who has extensively studied coyotes living in and around Chicago, said coyotes have been in the area for the past decade. He said the latest data shows there are at least 2,000 coyotes in Cook County, where Chicago is located.

"That's their territory. They live there," Gehrt, who works at Ohio State University, said of the unusual pair, which he said were born and raised in the city and likely live in Wrigleyville.

Byington had just a few minutes to snap his nighttime photos before the animals moved on. But he said the coyotes appeared unfazed by the honking horns and packed sidewalks. One even stepped onto a street as cabs and cars crawled by in traffic as if the coyote was "trying to figure out, do they want to eat? Or do they want to get a drink?" he said.

Byington didn't think much of his photos until he posted them online. But they started gathering attention after getting picked up on Facebook by the Cubby Bear bar where he had been working that night.

Chicago Cubs fan Dan Michaels reasons that the coyotes were likely fellow fans of the lackluster team and simply mixed up their calendars.

"My personal theory is they don't know that the Cubs play in summer," he said Wednesday as he walked by the stadium. "They picked the wrong season to wait out by the game."

Can Reality TV Boost Self-Control?Flipping through one of Psychology Today?s recent issues, my eyes focused on a short article ?Just Give In. Five Indulgences that actually boost self-control? by Kelly McGonigal, Ph.D.

What are these five indulgences? Personally, I was hoping chocolate was on the list. (Unfortunately, it?s not!) McGonigal narrows it down to these five things: a single espresso, an afternoon nap, a snack, YouTube and reality television.

The author writes that ?Willpower diminishes as the day wears on, but anything that reduces stress, boosts your mood, or recharges your energy can also reboot your self-control.?

Even ?reality TV??

Last night, after watching various contestants on a popular reality show sing for 90 minutes, I felt like falling asleep. Willpower? I must be watching the wrong program. Survivor might give me some energy: watching people trudge through swamps and sleep in tents, screaming at each other and falling in ?love? the next episode.

I kept reading, hopeful that the small paragraph devoted to the indulgence of reality television would explain our culture?s obsession with watching other people. People on a screen and not, say, upstairs in a different room or a phone call away.

?Willpower is contagious,? the author continues. ?Many reality shows feature people working hard to overcome obstacles as they lose weight, face their fears, or organize their clutter.?

I paused reading and recalled a past co-worker who spent, unfortunately, at least 30 minutes every single day talking about her favorite show, The Biggest Loser. We worked side-by-side and so this was particularly irritating, but even more so was the fact that she spent time talking to me ? and anyone else who would listen ? about her diets. She had a new diet every week and none of them worked.

I guess the show?s ?willpower? did not translate to her lifestyle. Sort of like how I can?t sing a note despite watching all of these silly singing shows.

The article concludes with ?You can ?catch? extra self-control just by watching someone pursue a goal.? Hmm. While the author of this article believes that television can boost willpower and productivity, I disagree. Watching television is a sedentary activity and it uses very few brain cells ? feeding your dog uses more. [Ed. - Although it may be a common popular perception, there's also little research that supports the idea that reality TV shows can help a person with their own willpower or act as a personal motivator.]

So, what?s the verdict? The reality is that watching reality television is a form of escape. I doubt it promotes ?self-control? (am I the only one who eats dinner while watching television?) but it has ingrained its way into our culture.

That aside, watching the presidential debates far exceeds the drama I have witnessed in any episode of Survivor. In the end, ?real life? is reality.

Acne in women can signal hormone problems - TODAY Health

Acne in adult women is common, but in some cases, it can signal an underlying hormonal disorder, experts say.

Identifying such cases is important, not only to better treat women's acne, but to correct hormone imbalances that can lead to other health problems, said Dr. Kanade Shinkai, a dermatologist at the University of California, San Francisco School of Medicine, who specializes in acne treatments.

However, doctors often simply give women prescriptions for acne treatments, such as antibiotics, which typically don't work for acne caused by hormone imbalances, Shinkai said

Women who visit the dermatologist for acne and who experience signs of a hormonal disorder, such as irregular periods, should discuss their symptoms with their doctors, Shinkai said.

In addition, it's important for dermatologists to conduct a thorough medical history of their patients so that they can identify hormonal disorders that manifest as acne, she said.

Acne in adult women is often hard to treat. One study showed that 80 percent of women prescribed antibiotics for acne receive no benefits from the drugs, Shinkai said.

In these cases, the acne may stem from normal hormonal changes, such as those that occur around the menstrual cycle, or from a true hormonal disorder. For many of these women, medications that target hormones, rather than skin bacteria, would provide the most help, Shinkai said.

Women with acne may have elevated levels of male hormones such as testosterone, Shinkai said. Symptoms of this type of hormonal imbalance include hair growth on the face, a deepening of the voice, increased muscle mass and decreased breast size.

The most common cause of such a hormonal change is a condition called polycystic ovarian syndrome, or PCOS. Women suspected of having a hormonal disorder can be diagnosed by having a doctor check their blood for elevated levels of male hormones, or in the case of PCOS, an ultrasound of the ovaries.

If a woman has PCOS or another type ofhormone imbalance, treating the underlying hormonal problem will help with the acne, Shinkai said.

Shinkai typically prescribes birth control pills to patients with PCOS, to lower their levels of male hormones. If those pills don't work by themselves, Shinkai adds another drug called spironolactone.

If left untreated, the hormone imbalances associated with PCOS may increase a woman's risk of cancer, as well as diabetes and obesity, which bring their own sets of medical problems, Shinkai said.

Birth control and spironolactone can also work very well against acne even in women with no hormonal disorder, Shinkai said.

Experts exhume remains of Palestinian leader

(AP) ? The remains of the late Palestinian leader Yasser Arafat were exhumed from his grave on Tuesday so international forensic experts could search for additional clues to his death, Palestinian officials said.

The remains were taken from the massive mausoleum in the West Bank city of Ramallah where Arafat was buried and moved to a nearby mosque so Palestinian doctors could take samples from his bones, the officials said. Under Islam, only Muslims can handle a Muslim's remains.

The samples will be handed over to French, Swiss and Russian experts who have flown in for the exhumation and who will examine them in their home countries, the officials said. Earlier, samples were also taken from Arafat's bedroom, office and personal belongings, they said.

The Palestinian officials spoke on condition of anonymity because they were not authorized to discuss the matter with the media.

The new investigation into Arafat's death was sparked earlier this year by the discovery of a lethal radioactive substance, polonium, on clothing said to be his.

Arafat died in November 2004 in a French military hospital, a month after suddenly falling ill. While the immediate cause of death was a stroke, the underlying source of an illness he suffered in his final weeks has never been clear, leading to persistent speculation in the Arab world that Israel poisoned him. Israel has denied such allegations.

The exhumation might not resolve the mystery. Polonium-210 decomposes rapidly, and some experts say it is not clear whether any remaining samples will be sufficient for testing.

(AP) ? Thousands flocked to Cairo's central Tahrir square on Tuesday for a protest against Egypt's president in a significant test of whether the opposition can rally the street behind it in a confrontation aimed at forcing the Islamist leader to rescind decrees that granted him near absolute powers.

Waving Egypt's red, white and black flags and chanting slogans against President Mohammed Morsi and his Muslim Brotherhood, the protesters joined several hundreds who have been camping out at the square since Friday demanding the decrees be revoked.

Even as the crowds swelled, clashes erupted nearby between several hundred young protesters throwing stones and police firing tear gas on a street off Tahrir leading to the U.S. Embassy. Clashes have been taking place at the site for several days fueled by anger over police abuses, separately from the crisis over Morsi.

The president's declaration last week of new powers for himself has energized and ? to a degree unified ? the mostly liberal and secular opposition after months of divisions and uncertainty while Islamists from the Muslim Brotherhood and other groups rose to dominate the political landscape.

The turnout for Tuesday's protest call is key to whether the opposition can keep a movement going against Morsi. While the edicts last week sparked the protests, they have also been fueled by anger over what critics see as the Brotherhood's monopolizing of power after its election victories the past year for parliament and the presidency.

"We want to change this whole setting. The Brotherhood hijacked the revolution," said Rafat Magdi, an engineer who was among a crowd of around 2,000 marching from the Cairo district of Shubra to Tahrir to join the rally. "People woke up by his mistakes, and any new elections they will get no votes."

Morsi's decrees, issued Thursday, placed him above any kind of oversight, including that of the courts, until a new constitution is adopted and parliamentary elections are held ? a timeline that stretches to mid-2013.

The opposition says the decrees give Morsi near dictatorial powers by neutralizing the judiciary at a time when he already holds executive and legislative powers. Leading judges have also denounced the measures.

Morsi says the decrees are necessary to protect the "revolution" and the nation's transition to democratic rule. His declaration made all his decisions immune to judicial review and banned the courts from dissolving the upper house of parliament and an assembly writing the new constitution, both of which are dominated by Islamists. The decree also gave Morsi sweeping authority to stop any "threats" to the revolution.

By early afternoon, nearly 20,000 people were at Tahrir, birthplace of the 18-day popular uprising that toppled President Hosni Mubarak's authoritarian regime nearly two years ago. Security was tightened across Cairo, with police checking ID papers and searching cars coming into the center, but there was no sign of protesters being stopped from reaching Tahrir.

A chaotic city of some 18 million people, Cairo's traffic was uncharacteristically light on Tuesday, with many businesses and government offices closing shop early in anticipation of possible unrest.

A new banner in the square proclaimed, "The Brotherhood stole the country" and one protester, Mahmoud Youssef, said: "We are here to bring down the constitutional declaration issued by Morsi." Many chanted the iconic slogans "the people want to bring down the regime."

Several thousand lawyers meanwhile gathered outside their union building in downtown Cairo ahead of their march to Tahrir to join protesters there. "Leave, leave," they chanted, addressing Morsi, who narrowly won elections in June to become the country's first freely elected civilian president.

In the Mediterranean city of Alexandria, some 3,000 anti-Morsi protesters gathered outside the main court at the center of the ancient city.

Morsi's supporters canceled a massive rally they had planned for Tuesday, citing the need to "defuse tension" after a series of clashes between the two camps since the decrees were issued Thursday.

But a Brotherhood spokesman said demonstrations supporting the president could go ahead outside the capital and that supporters would form human chains in some provinces to protect Brotherhood offices. Morsi's supporters say more than a dozen of their offices have been ransacked or set ablaze since Friday. Some 5,000 demonstrated in the southern city of Assiut in support of Morsi's decrees, according to witnesses there.

On Monday, Morsi met with the nation's top judges and tried to win their acceptance of his decrees. But the move was dismissed by many in the opposition and the judiciary as providing no real concessions.

Presidential spokesman Yasser Ali, said Morsi told the judges that he acted within his rights as the nation's sole source of legislation, assuring them that the decrees were temporary and did not in any way infringe on the judiciary. He underlined repeatedly that the president had no plans to change or amend his decrees.

According to a presidential statement late Monday, Morsi told the judges that his decree meant that any decisions he makes on "issues of sovereignty" are immune from judicial review.

The vaguely worded statement did not define those issues, but they were widely interpreted to cover declaration of war, imposition of martial law, breaking diplomatic relations with a foreign nation or dismissing a Cabinet. Morsi's original edict, however, explicitly gives immunity to all his decisions and there was no sign it had been changed.

Monday's presidential statement Monday did not touch on the immunity that Morsi gave the constitutional assembly or the upper chamber of parliament, known as the Shura Council. It also did not affect the edict that the president can take any measures he sees as necessary to stop threats to the revolution, stability or public institutions. Many see that edict as granting Morsi unlimited emergency powers.

The Shura Council does not have lawmaking authorities but, in the absence of the more powerful lower chamber, the People's Assembly, it is the only popularly elected, national body where the Brotherhood and other Islamists have a majority. The People's Assembly was dissolved by a court ruling in June.

Rights lawyers and activists, however, dismissed Morsi's assurances as an attempt to defuse the crisis without offering concrete concessions.

One of the lawyers, Ahmed Ragheb, described the presidential statement and Ali's comments as "playing with words."

"This is not what Egyptians are objecting to and protesting about," he said. "If the president wanted to resolve the crisis, there should be an amendment to his constitutional declaration."

On Tuesday, the influential Judges' Club, a sort of union led by an outspoken Morsi critic, vowed in a statement to escalate its resistance to the decrees. The statement was carried by Egypt's official MENA news agency, which also reported that judges and prosecutors continued their strike for the third day Tuesday, leaving many courtrooms empty across the nation.

Fertility: Putrescine water may be fountain of youth for human eggs

ScienceDaily (Nov. 26, 2012) ? An Ottawa scientist has discovered a critical reason why women experience fertility problems as they get older. The breakthrough by Dr. John? Liu, a senior scientist at the Ottawa Hospital Research Institute and professor at the University of Ottawa, also points to a simple solution that could increase the viability of egg cells for women in their late 30s and older -- putrescine water.

In an online editorial published by Aging based on his recently published findings, Liu outlines how a simple program of drinking water or taking a pill that contains the naturally occurring compound putrescine could reduce the rate at which middle-aged women produce eggs with the incorrect number of chromosomes, the leading cause of reduced fertility and increases in miscarriages and congenital birth defects.

Putrescine is naturally produced in mammals by an enzyme called ornithine decarboxylase, or ODC, and is easily absorbed and cleared by the body. In female mammals, ODC levels are known to rise during ovulation, when the egg cell matures and is released from the ovary. Dr. Liu has shown that ODC levels rise very little in older females. He has also shown that inhibiting ODC levels in young mice leads to an increase in egg cells with chromosomal defects.

Taking this a step further, Dr. Liu's team gave older mice putrescine water in the period immediately leading up to and during ovulation, and found that it reduced the incidence of defective eggs by more than 50%.

This is a remarkable outcome for such a simple approach," says Dr. Liu. "However, we could not have imagined this without first understanding the role that ODC and putrescine play in maintaining the chromosomal integrity of egg cells. While there is work to be done before it can be approved for clinical use, we feel this approach could be used for natural conception as well as in vitro fertilization."

Although promising, a putrescine pill is still a long way from being available on the market. Putrescine is toxic to the fetus if it is administered after conception, which makes timing, dosage and monitoring critical. Accordingly, this approach must go through the appropriate stages of testing to determine its clinical safety and effectiveness in humans.

The full paper, "Deficiency of ovarian ornithine decarboxylase contributes to aging-related egg aneuploidy in mice," was recently accepted for publication online ahead of print by Aging Cell. The editorial was published online by Aging on November 23. Yong Tao of the Ottawa Hospital Research Institute and the University of Ottawa is co-author on both publications.

Funding for this research was provided by a Discovery Grant from the Natural Sciences and Engineering Research Council of Canada and a partial scholarship from the Canadian Institutes of Health Research's Training Program in Reproduction, Early Development, and the Impact on Health.

In a NYT Op-Ed today, Warren Buffett argued that the rich should pay a certain minimum tax, and he explained to activist Grover Norquist that it's preposterous to think that businessmen would forgo profitable deals merely because the rate of tax on the profits would go up.

But why do people listen to Buffett on taxes?

Basically because he's a rich, successful guy (which is why a lot of people are listened to on a lot of subjects).

To that end, economist and former Romney advisor Greg Mankiw has a short post talking about Buffett as a master of "tax avoidance," wherein he lists four things Buffett does to avoid paying taxes.

  • Berkshire never pays a dividend (so the jump in dividend tax hikes don't effect him).
  • Berkshire only trades long-term? (so short-term cap gains, which are taxed at income tax rates don't effect him).
  • He's giving most of his money away to charity.
  • His children won't pay income taxes on any assets that are bequeathed to them, so an income tax hike doesn't affect them.

None of these are wrong or illegal or anything. And giving your money away to charity is admirable. So the point isn't that Buffett is doing anything wrong, but that he's advocating policies which will have very little effect on him. So if we're going to listen to him because of who he is, it's not preposterous to note how little of an effect rule changes would have on him.

Other folks, in the past, have charged that Buffett specifically benefits from the tax proposals he backs.

Tim Carney of the Washington Examiner has pointed out:

Buffett regularly lobbies for higher estate taxes. He also has repeatedly bought up family businesses forced to sell because the heirs? death-tax bill exceeded the business?s liquid assets. He owns life insurance companies that rely on the death tax in order to sell their estate-planning businesses.

All that being said, the arguments against raising taxes marginally on the rich (assuming a deficit reduction deal is warranted) tend to be pretty poor.
